Barritt Reagent A (for VP test) 100ml About

HiMedia's Barritt Reagent A is a light yellow colored solution used with Barritt Reagent B in the Voges-Proskauer (VP) test to detect acetoin, a neutral end product of glucose fermentation by bacteria. A positive VP test, indicated by the formation of a cherry-red color after adding both Barritt reagents, helps identify bacteria like Enterobacter aerogenes. This reagent is a for in-vitro use, stored at 15-25C, and should not be exposed to skin or eyes.
 Purpose and Use
  • Voges-Proskauer Test: Barritt Reagent A is part of a two-reagent system used in the VP test. 
  • Acetoin Detection: The test identifies bacteria that ferment glucose to produce neutral, non-acidic end products, primarily acetoin (acetyl methyl carbinol). 
  • Bacterial Identification: The VP test is particularly useful for identifying members of the Enterobacteriaceae family. 
Procedure
  1. Inoculate a bacterial culture on MR-VP medium and incubate it for 24-48 hours. 
  2. Add Barritt Reagent A, followed by Barritt Reagent B (a potassium hydroxide solution). 
  3. Observe for the development of a cherry-red color within 5-10 minutes. 
Interpretation of Results 
  • Positive Test: A cherry-red color indicates the presence of acetoin and a positive VP test.
  • Negative Test: No red color formation signifies a negative VP test.
Storage and Safety
  • Storage: Store Barritt Reagent A at 15-25C. 
  • Safety: It is a combustible reagent and should be handled with care. Use appropriate personal protective equipment, such as gloves and eye protection, during handling.
