This enzyme plays a crucial role in the electron transport chain and is a key factor in identifying certain bacteria, particularly pseudomonads and other oxidase-positive species, according to the American Society for Microbiology. Detailed Description:
  • Purpose:The reagent facilitates the oxidase test, a microbiological procedure used to differentiate bacteria based on their ability to produce cytochrome c oxidase. 
  • Composition:The reagent typically contains N,N,N,N-tetramethyl-p-phenylenediamine dihydrochloride (also known as tetramethyl-p-phenylenediamine) as the active component. This compound is a colorless reagent in its reduced form. 
  • Mechanism:When cytochrome c oxidase is present in a bacterial culture, it oxidizes the tetramethyl-p-phenylenediamine, causing it to change color from colorless to a dark blue or purple, indicating a positive result. 
  • Application:The oxidase test is used to help distinguish between different bacterial species, particularly Neisseria, Moraxella, Campylobacter, and Pasteurella, which are generally oxidase-positive, and other bacterial groups. 
  • Storage:Oxidase reagents are typically stored in a cool place, such as a refrigerator (2 to 8C) to maintain their stability and prevent degradation. 
  • Procedure:In the oxidase test, a small amount of the Gordon-McLeod reagent is applied to a filter paper or other suitable surface, and then bacterial colonies are smeared onto the paper. A positive result is indicated by the appearance of a blue or purple color within a short time (e.g., 10 seconds).
