The HiMedia Gram Stain Kit (like K001/K001L) provides reagents (Crystal Violet, Gram's Iodine, Decolorizer, Safranin) to perform the differential Gram stain, essential for classifying bacteria by cell wall type (thick peptidoglycan retains purple/Gram-positive; thin layer loses stain and becomes pink/Gram-negative) and guiding diagnosis, following the classic staining principle where the iodine mordant fixes the violet stain, decolorizer washes it from Gram-negatives, and safranin counterstains them. Key Components & Function:
  • Gram Crystal Violet: The primary stain that colors all bacteria purple initially.
  • Gram Iodine: A mordant that forms a complex with the crystal violet, making it stick better to Gram-positive cells.
  • Gram Decolorizer (Alcohol/Acetone): Washes the stain from Gram-negative cells with thin walls but leaves it in Gram-positive cells with thick walls.
  • Gram Safranin: A counterstain that colors the now-clear Gram-negative cells pink/red. 
Principle:
  1. Stain with Crystal Violet: All cells purple.
  2. Apply Iodine: Forms large complexes.
  3. Decolorize: Gram-positives stay purple; Gram-negatives become colorless.
  4. Counterstain with Safranin: Gram-positives remain purple; Gram-negatives turn pink/red.
