• Chromogenic substrates: Detect the activity of -D-galactosidase and -D-glucuronidase
• Peptone special: Provides nutrients for organisms to grow
• Sodium pyruvate: Provides nutrients for organisms to grow
• Phosphates: Buffer the medium
• Sodium lauryl sulfate: Inhibits gram-positive organisms
• L-Tryptophan: Improves the indole reaction

• Coliforms produce the enzyme -galactosidase, which cleaves a chromogen and turns coliform colonies salmon red
• E. coli produces the enzyme -glucuronidase, which cleaves X-glucuronide and turns E. coli colonies dark blue to violet

• Store in a tightly closed container between 1525C
• Use before the expiration date
• Dispose of used or unusable preparations by autoclaving or incineration
