HiCrome Chromogenic Coliform Agar (CCA Agar) isa selective medium used to detect coliforms and Escherichia coli in water and food samples.It contains chromogenic substrates that detect the enzymes -galactosidase and -glucuronidase.What it detects:
  • ColiformsProduce pink colonies due to the cleavage of one chromogen by the enzyme -galactosidase
  • Escherichia coliProduce dark blue to violet colonies due to the cleavage of both chromogens by the enzymes -galactosidase and -glucuronidase
How it works:
  • The medium contains nutrients like peptone special and sodium pyruvate to help organisms grow
  • Sodium lauryl sulfate inhibits gram-positive organisms
  • L-Tryptophan improves the indole reaction, which increases detection reliability
How to use:
  • To confirm E. coli, add a drop of Kovacs reagent to the dark-blue to violet colony
  • If the color turns cherry-red, the reaction is positive
Storage:
  • Store in a tightly closed container between 15-25C
  • Use before the expiry date on the label
  • Dispose of used or unusable preparations by autoclaving or incineration
