MacConkey Agar (MAC) is a selective and differential culture medium used in microbiology, primarily for isolating and differentiating gram-negative bacteria.It is named after bacteriologist Alfred T. MacConkey.Key Features of MacConkey Agar (Himedia):
  • Selective:It inhibits the growth of gram-positive bacteria, allowing only gram-negative bacteria to grow.
  • Differential:It differentiates lactose-fermenting bacteria from non-lactose-fermenting bacteria.
  • Components:It typically contains lactose, bile salts, neutral red, and crystal violet.
  • Lactose Fermentation:Lactose-fermenting bacteria produce acid, which lowers the pH, causing the neutral red indicator to turn pink or red, and the colonies appear pink or red.
  • Non-Lactose Fermenters:Bacteria that do not ferment lactose do not produce acid, and the colonies remain colorless or the agar surrounding them remains a pale yellow.
  • Application:Commonly used for isolating and identifying bacteria from various sources, including clinical samples and food.
