They are commonly used to differentiate between bacteria, particularly to identify oxidase-positive bacteria like Neisseria, Alcaligenes, Aeromonas, Vibrio, Campylobacter, and Pseudomonas. Key Features and Usage:

Reagent:The discs are impregnated with a mixture of N,N-dimethyl-p-phenylenediamine oxalate, ascorbic acid, and -naphthol.
Detection:The reagent acts as an electron donor. When a colony of oxidase-positive bacteria is placed on the disc, the oxidase enzyme oxidizes the reagent, causing a color change, typically a purplish-blue.
Interpretation:A color change indicates a positive result, signifying the presence of oxidase enzyme. No color change is considered a negative result.
