Properties Purpose: PCA is a microbiological growth medium used to assess or monitor the total viable bacterial growth in a sample. - Composition: * Enzymatic Digest of Casein/Tryptone (5.0 g/L) * Yeast Extract (2.5 g/L) * Glucose (1.0 g/L) * Agar (15.0 g/L) * Method: * Not selective; supports the growth of aerobic bacteria. * Inoculum added to molten agar, which is then poured into a petri dish. * Incubated for about 3 days at 20-30 C. * Results: * Bacteria counted as colony-forming units per gram (CFU/g) in solid samples or CFU/mL in liquid samples. * Benefits of PCA: * Easy to perform. * Larger sample volume than surface spread method. * No need for pre-drying agar surface. * Detects lower microbiological concentrations. * Useful for assessing bacterial contamination in foods. * Isolating Colonies: * Once prepared, colonies grow on PCA plates. * Streak plate method helps identify unknown microbes. * CFU allows correct description of cell density
