Chromogenic coliform medium with a sterile membrane filter is a specialized microbiological test kit used to detect and count E. coli and total coliforms in water samples
Overview and Purpose
This method is efficient and rapid, making it suitable for water quality analysis.
by combining membrane filtration with chromogenic (color-producing) agents. It allows labs to identify target bacteria based on distinct colony colors without needing complex secondary tests
How It Works
  • Filtration: The test sample should be filtered through a sterile membrane filter having pore size of 0.22 / 0.45. to trap bacteria on the filter surface.
  • Color Differentiation: The medium contains specific substrates cleaved by enzymes (like -glucuronidase) found in E. coli and other coliforms, causing specific color reactions in the growing colonies.
  • Pad Rehydration: Rehydrate the nutrient pad with 2.0-2.5 ml sterile distilled / purified water. before placing the processed filter on top.
  • Test Directions
    1. Pass the water sample through the provided sterile membrane filter unit.
  • Wet the absorbent nutrient pad with sterile water.
  • Use sterile forceps to transfer the membrane filter onto the wet pad, filter-side up.
  • Seal and incubate the plate according to product specifications (commonly around 35C to 37C for 2124 hours).
  • Count colored colonies quantitatively as CFU/ml and read qualitative growth results.
