HiMedia's Cooked Meat Medium (R.C.Medium) is a rich, reducing medium used to cultivate fastidious anaerobes, especially pathogenic Clostridia, from clinical, food, and water samples, providing nutrients from beef heart and a reducing environment with glutathione for obligate anaerobes, indicated by turbidity or meat disintegration, and is also for maintaining stock cultures. Key Features & Purpose:
  • Cultivation: Ideal for Clostridium species (like C. welchii) and other aerobes/anaerobes.
  • Reducing Environment: Contains cooked beef heart and glutathione, which create low oxidation-reduction potential (redox) for anaerobes to thrive.
  • Nutrient Rich: Beef heart provides essential amino acids and growth factors.
  • Stock Culture Maintenance: Used to preserve bacterial cultures.
  • Proteolytic Activity: Helps determine if anaerobes can break down protein (indicated by meat blackening/disintegration).

How it Works:
  1. Meat Particles: The cooked meat provides a surface and nutrients, while its denatured proteins offer available sulfhydryl groups, creating the necessary reducing conditions.
  2. Glutathione: Acts as a powerful reducing agent, scavenging oxygen and allowing strict anaerobes to grow.
  3. Dextrose (in some formulations): Allows for rapid growth and detection of anaerobes. 
Usage:
  • Prepare by suspending powder in distilled water, mixing, letting it wet, and autoclaving.
  • Best used fresh; if stored, reheat gently (steam/boil) and cool without agitation before inoculation.
  • Growth appears as turbidity or gas bubbles; blackening/disintegration signals proteolysis.
