About Fast Green FCF For Microscopy

Fast Green FCF (C.I. 42053) is a synthetic green dye, primarily used in microscopy as a histological stain for connective tissues and as a counterstain. It's also employed in various trichrome staining methods, such as Masson's trichrome, and as a substitute for Light Green SF in Papanicolaou staining. Furthermore, it's used in protein staining techniques like SDS-PAGE and IEF (Isoelectric Focusing). Key Uses:
  • Histology and Cytology:
    • Stains connective tissues, providing a green contrast to other stained structures.
    • Often used as a counterstain in methods that stain structures blue, purple, or red.
    • Improves the visualization of extracellular matrix (ECM) in microscopy.
  • Trichrome Staining:
    • Used in Masson's trichrome and Gomori's trichrome methods.
  • Protein Staining:
    • Suitable for protein staining in electrophoresis, particularly SDS-PAGE and IEF.
    • Offers linear staining over a wider range of protein concentrations than some other dyes according to Ennore India Chemicals.
  • Histones:
    • Can be used as a quantitative stain for histones at alkaline pH after acid extraction of DNA according to Wikipedia.
Advantages:
  • Brilliant Color: Fast Green FCF provides a more brilliant and less fading color than Light Green SF, making it a preferred replacement in some staining techniques.
  • Sensitivity: It offers good sensitivity for protein detection in electrophoresis.
  • Quantitative Staining: Its ability to stain histones quantitatively is valuable in certain research applications.
Considerations:
  • Solubility:Fast Green FCF is highly soluble in water but less so in alcoholic solutions.
  • Safety:It's important to handle Fast Green FCF according to established safety guidelines, as it can be a potential health hazard.
  • Disposal:Used solutions should be disposed of as special waste according to national guidelines.
