About Fluorescein Diacetate 5g Sigma-Aldrich

Fluorescein diacetate (FDA) is a nonfluorescent, lipid-soluble dye used to assess cell viability and membrane integrity, as living cells with active esterase enzymes hydrolyze FDA into the green fluorescent compound fluorescein. Dead or non-viable cells cannot perform this hydrolysis, so only live cells accumulate the green fluorescence. This process makes FDA a vital stain that can be combined with other stains like propidium iodide (PI) for a two-color analysis of viable (green) and non-viable (red) cells. How it works
  1. 1. Entry into the cell:The nonfluorescent FDA, being lipid-soluble, can readily cross the cell membrane of live cells. 
  2. 2. Enzymatic hydrolysis:Inside the cell, nonspecific intracellular esterase enzymes cleave the diacetate group from the FDA molecule. 
  3. 3. Fluorescence activation:This cleavage releases free fluorescein, a highly green fluorescent compound that accumulates within the cell. 
  4. 4. Viability assessment:The presence of green fluorescence indicates that the cell is alive and has the enzymatic activity to convert FDA into fluorescein.
