About Kovac's Indole Reagent 100ml Himedia

HiMedia product R008 is Kovac's Indole Reagent, a biochemical reagent used in microbiology to perform the indole test. This test helps differentiate microorganisms, particularly members of the family Enterobacteriaceae, based on their ability to produce indole from the amino acid tryptophan. Product Description
  • Intended Use: To detect the production of indole by bacteria that possess the enzyme tryptophanase.
  • Principle: Certain bacteria can break down tryptophan into indole, pyruvic acid, and ammonia. Kovac's reagent reacts with the indole to produce a colored product.
  • Composition: The reagent typically contains p-dimethylaminobenzaldehyde (p-DMAB), amyl alcohol (or isoamyl alcohol), and concentrated hydrochloric acid.
  • Appearance: A clear, often yellowish or amber liquid solution.
  • Pack Size: Commonly available in 100 mL glass bottles.
  • Storage & Handling: It should be stored in a cool, dark place, away from ignition sources, as it is a flammable liquid. 
Test Procedure and ResultsTo perform the test using HiMedia R008:
  1. Grow the test organism in a medium rich in tryptophan, such as Tryptone Broth (M463), for 24-48 hours.
  2. Add a few drops (0.2-1 ml depending on the protocol) of Kovac's reagent (R008) to the culture medium.
  3. Observe for color development at the interface of the medium and the reagent layer. 
  • Positive Result: The rapid formation of a pink to red-colored ring at the top interface indicates a positive reaction (presence of indole) (e.g., Escherichia coli).
  • Negative Result: The reagent layer remains yellow or a cloudy yellow-brown, indicating a negative reaction (absence of indole) (e.g., Enterobacter aerogenes
