About Tryptophan Medium Himedia 500g

HiMedia's Tryptophan Medium is a microbiological broth used for the Indole Production Test, helping differentiate bacteria like E. coli by detecting if they can break down the amino acid tryptophan into indole, which is then identified with Kovac's reagent (red ring). It's a yellowish powder that forms a clear, light yellow solution with a pH of approximately 7.5, providing essential nutrients and the substrate (tryptophan) for microbial growth and testing. Key Characteristics:
  • Purpose: Detects indole production, a key feature in differentiating bacterial species (e.g., coliforms).
  • Appearance (Powder): Cream to yellow, free-flowing, homogeneous powder, sometimes with a slight greenish tinge.
  • Appearance (Prepared): Clear, light yellow solution.
  • pH: ~7.5 0.2 (for a 1.6% w/v solution).
  • Principle: Microorganisms possessing the enzyme tryptophanase hydrolyze tryptophan; indole is detected by adding Kovac's or Ehrlich's reagent, forming a red color.
  • Variations: Includes HiVeg versions (using vegetable peptones for BSE/TSE safety) and formulations for specific standards (like ISO). 
How it Works (Indole Test):
  1. Growth: Bacteria grow in the medium, utilizing nutrients and the tryptophan.
  2. Enzymatic Action: If the organism has tryptophanase, it breaks down tryptophan to produce indole.
  3. Detection: Kovac's reagent is added, reacting with indole to form a red layer, indicating a positive test.
