Xylose Lysine Deoxycholate Agar 500g Himedia About

Xylose Lysine Deoxycholate Agar (XLD Agar) is a selective and differential medium used for isolating and identifying Salmonella and Shigella bacteria, particularly from clinical samples and food. It's characterized by its pink color, a free-flowing, homogeneous powder. XLD Agar relies on sodium deoxycholate as a selective agent to inhibit Gram-positive microorganisms, and a combination of xylose, lysine, and other components for differential growth and identification of specific bacteria. Here's a more detailed breakdown:Selective Features:
  • Sodium Deoxycholate: This component inhibits the growth of most Gram-positive bacteria, ensuring that only Gram-negative enteric bacteria like Salmonella and Shigella can thrive.
Differential Features:
  • Xylose:Salmonella and Shigella utilize xylose, a sugar, as a carbon source, leading to the production of acid, which changes the pH indicator (phenol red) from red to yellow, allowing for differentiation.
  • Lysine:Lysine is used as a carbon and nitrogen source. After xylose is exhausted, Salmonella will decarboxylate lysine, producing an alkaline pH, which mimics the Shigella reaction and can help distinguish them.
  • Hydrogen Sulfide Production:Some Salmonella species can produce hydrogen sulfide (H2S) from the thiosulfate in the medium, resulting in black colonies, a characteristic feature for differentiation.
  • Lactose, Sucrose, and Phenol Red:These components, along with lysine and xylose, are part of the differential system, allowing for the identification of bacteria based on their fermentation patterns and hydrogen sulfide production.
Colony Appearance:
  • Salmonella: Red to pink colonies, some with black centers due to hydrogen sulfide production.
  • Shigella: Red colonies, as they do not ferment xylose or produce hydrogen sulfide.
  • Coliform Bacteria (e.g., E. coli): Yellow or orange colonies.
Uses:
  • Isolation of Salmonella and Shigella:XLD Agar is used to isolate these bacteria from various sources, including stool samples, food, and clinical specimens.
  • Identification of Enteric Pathogens:It helps differentiate between different enteric pathogens, including Salmonella, Shigella, and other Gram-negative bacteria.
  • Pharmaceutical and Food Testing:XLD Agar is also used in pharmaceutical and food testing to detect the presence of Salmonella and other bacteria.
