About Mannitol Salt Agar 500g Himedia

Mannitol Salt Agar (MSA) is a type of growth medium used in microbiology that is both selective and differential. It's primarily used to isolate and identify Staphylococcus aureus from other bacteria, especially those that can tolerate high salt concentrations. MSA contains a high salt concentration (7.5%), which inhibits the growth of most bacteria, making it selective for halotolerant organisms like staphylococci. It also differentiates Staphylococcus aureus from other staphylococci based on their ability to ferment mannitol, a sugar alcohol, which is indicated by a color change in the medium. Here's a more detailed explanation:
  • Selective:MSA selects for Staphylococcus species (and other halotolerant bacteria) due to its high salt concentration (7.5% NaCl). This inhibits the growth of most Gram-negative bacteria and many Gram-positive bacteria except those that can tolerate high salt. 
  • Differential:MSA distinguishes between Staphylococcus aureus and other staphylococci based on their ability to ferment mannitol. Staphylococcus aureus can ferment mannitol, producing acidic byproducts that lower the pH of the medium, causing the pH indicator phenol red to turn yellow. Other staphylococci that cannot ferment mannitol will grow with pink or red colonies. 
  • Other Components:Besides NaCl, MSA also contains beef extract and proteose peptone, which provide essential nutrients for bacterial growth. 
  • Purpose:MSA is commonly used in labs to identify presumptive Staphylococcus aureus isolates, particularly in clinical samples or food samples. 
  • Growth on MSA:
    • Staphylococcus aureus colonies will appear yellow or with a yellow halo due to mannitol fermentation. 
    • Other staphylococci will appear pink or red on the medium. 
    • Most other bacteria are inhibited by the high salt concentration.
