It's particularly useful for differentiating Proteus species from other enteric bacilli. Urease is an enzyme that some bacteria produce to break down urea, releasing ammonia and causing the medium to become alkaline. A positive result, indicated by a color change from yellow to pink or red due to the pH indicator phenol red, signifies urease production. Here's a more detailed breakdown:
  • Urease Test:This medium is used to test for the presence of urease, an enzyme that some bacteria produce to hydrolyze urea. 
  • Urea Utilization:Bacteria with urease produce ammonia from urea, which raises the pH of the medium and causes a color change in the indicator. 
  • Differentiation:Stuart's Urea Broth Base is helpful in distinguishing Proteus species, which are typically urease-positive, from other enteric bacteria like Salmonella and Shigella, which are often urease-negative.
